What is OS-01?
OS-01 is the name OneSkin uses for a synthetic peptide in its face, body and scalp products. In the company's main research paper, published in 2023, it appears as Peptide 14. OneSkin scientists screened hundreds of peptides on skin cells from a person with progeria, a rare premature-aging disease, looking for ones that reduced signs of cellular senescence. Peptide 14 was chosen from the top performers and was then tested on skin cells from older donors and in other aging models.
Senescent cells are cells that have stopped dividing but do not die. They release inflammatory signals that can affect nearby tissue, and they become more common with age. Compounds that target them are called senotherapeutics. OneSkin's researchers describe OS-01 as senomorphic, meaning it appears to change how senescent cells behave rather than killing them.
How does OS-01 work?
The exact mechanism is still being worked out. In the 2023 study, Peptide 14 reduced markers of senescence in skin cells without causing them to die. Gene and protein data pointed to effects on PP2A, an enzyme involved in keeping the genome stable, and on genes tied to DNA repair and the cell cycle. The authors proposed that it helps keep early senescent cells from becoming late senescent cells, which release more inflammatory signals. These findings come from lab-grown cells and donated skin samples, not living people.
What has research looked at?
- Senescence in skin samples
- In lab-grown skin models and donated human skin, Peptide 14 lowered senescence markers and, in one analysis, reduced a DNA methylation estimate of the skin's biological age.
- Comparison with retinol
- In donated skin samples, topical Peptide 14 thickened the outer skin layer without the irritation markers seen with retinol. This was a lab comparison, not a human trial.
- Body lotion trial
- A 12-week randomized, double-blind pilot in 60 women over age 60 compared an OS-01 lotion with a standard moisturizer. The company-run study reported improved barrier function in both groups, somewhat more with OS-01, plus better hydration and favorable changes in some blood markers of inflammation.
- Other skin studies
- OneSkin researchers have also published a lab and patch-test safety assessment and small studies of OS-01 on facial and under-eye skin.
- Hair and scalp
- Results for the company's scalp product have mainly been reported by OneSkin itself rather than in peer-reviewed journals.
OS-01 has more published data than many cosmetic peptides, but nearly all of it comes from OneSkin's own scientists. The human studies are small, short and limited to skin. There is no published evidence for effects on muscle, heart, body weight or whole-body energy use.
OS-01 and AMPK
OS-01 is sometimes described online as an exercise mimic that switches on the energy sensor AMPK and builds new mitochondria. OneSkin's published studies haven't looked at it that way: they focus on topical use and on senescent skin cells. The AMPK claims closely resemble published work on a different experimental compound, O304, a small molecule rather than a peptide, studied in aged mice. Until studies of OS-01 itself test those energy effects, they remain unconfirmed for this peptide.
Side effects and cautions
In OneSkin's published studies, topical OS-01 products were generally well tolerated, though a small number of people stopped because of skin reactions. Safety data come from short studies of skincare products, so long-term effects, and the effects of any other form of use, are unknown. As with any new skin product, people with sensitive skin or eczema may react. Anyone with a skin condition, or who is pregnant, should talk to a qualified clinician.
- Skin irritation
- In the body lotion trial, three people using OS-01 dropped out because of rash, pimples or dry patches, which the authors said could have come from the lotion or the cleanser used.
- Other forms of use
- There is no published human safety data for OS-01 taken by mouth or by injection.
- Claims beyond the data
- Claims about fat loss, muscle, heart or brain effects are not supported by published OS-01 research.
